Background & aim:Excessive production of free radicals induces apoptosis of osteochondral cells and degradation of the extracellular matrix, ultimately leading to osteochondral defects. Given that the essential oil of Pelargonium graveolens (rose geranium) contains antioxidant constituents, the present study aimed to determine the effects of rose-geranium leaf essential oil on the expression levels of glutathione (GSH), malondialdehyde (MDA), superoxide dismutase (SOD), and 8-hydroxy-2'-deoxyguanosine (8-OHdG) in collagenase-induced osteoarthritis of the rabbit knee joint. Methods: This experimental study was conducted in 2021 on 30 New Zealand white rabbits. After preparation of the plant essential oil, the rabbits were acclimatized to laboratory conditions for one month. Osteoarthritis was induced by intra-articular injection of collagenase into the right knee joints. Thirty days after OA induction, the rabbits received oral treatments of distilled water (positive control), rose-geranium leaf essential oil at 50 or 100 mg/kg, or celecoxib (0.3 mL of 0.4%), once daily for 30 days. At the end of weeks 4, 8, and 12, levels of GSH, MDA, SOD, and 8-OHdG were measured in peripheral blood and synovial fluid. Data were analyzed using one-way ANOVA, Tukey’s post hoc test, and t-tests. Results:In the essential-oil–treated group, GSH and SOD levels at weeks 8 and 12 were significantly higher than in the control group (p<0.05). Conversely, MDA and 8-OHdG levels indicated an increasing trend in the positive control group, whereas a significant reduction was observed in the treated groups (p<0.05). Conclusion: Rose-geranium leaf essential oil enhanced antioxidant activity by increasing GSH and SOD expression and reducing oxidative-stress markers (MDA and 8-OHdG). These findings suggest a potential protective role of this essential oil against oxidative stress–induced osteochondral damage.
